Summary of ScienceDome operations: I used it for my analysis of concretions. A quick test was performed on the concretion samples collected by Crew 291 using vinegar. 100 ml Vinegar was used for the test keeping safety in check. I used a hammer from RAM to crush and sieve the rock samples. Crew Biologist targeted PCR for ID, made up media, and inoculated them with samples from yesterday. He also performed another targeted PCR for yesterday’s samples. He also taught crew members (Bharti, Clare and me) the procedure of performing PCR for DNA identification on the samples collected from the field. Crew Artist, Green Hab Officer and Astronomer used it for their drawings.

Summary of Science Dome operations: In the morning the science dome was used to run one gel visualisation of the functional PCR test carried out yesterday on the DNA extracted from the "soil 2" sample and found that the DNA contains the large subunit gene hhyL of the group 1h [NiFe]-hydrogenases, potentially meaning that there are organisms present in the soil that perform hydrogen oxidation. In the afternoon non-targeted PCR run and gel visualisation of DNA extracted from media that appears to have grown microorganisms from previous inoculations, ultimately no DNA was found in the extracts.

Status on suit #6: I have replaced the battery and have completed the 1-hour test. It was 13.0 V and reduced to 11.9 V after 1 hour. Suit #6 is now operational.

Summary of ScienceDome operations: On EVA 9 in the morning two samples were taken, one from the soil in a crevice worn out of a larger rock, the other from a yellow goo (crew journalist’s description). In the afternoon DNA extraction was performed on the collected samples and non-targeted PCR was run on the previously extracted samples from yesterday; two water samples and two soil samples. As this report is being written the gel to visualize results is running. Our Crew Journalist Clare used it to clean rock specimens.

These were inoculated with 0.25 g of the two different types of soil samples collected yesterday on EVA 4 and placed in the incubator which is set at 37 C 99F to see if anything will grow. Bharti used the Science Dome for her literature review on her research project. Annalea used the Science Dome to make drawings of concretions for her project.

The voltage before EVA was 13.12 V and post EVA it was 9.15V. The fan running low might be due to the low voltage. I remember Crew 291 Engineer performed a 1-hour fan run test to see if the battery was faulty for a similar situation. Do I have permission to run that test on Suit #6? Need Advice.

Summary of Science Dome operations: Daniel used the Science Dome after sending the EVA team out, starting a non-targeted PCR run of extracted DNA from samples collected on EVA 4 on Sol 4. Towards the end of the run, he prepared an electrophoresis gel to visualize the results of the PCR and found DNA from soil that was collected from underneath a rock. An image was taken and annotated, further PCR’s will be performed to identify this DNA and investigate the presence of a range of biogeochemical genes. Annalea used it for her drawings on concretions.

Summary of ScienceDome operations: In the morning Daniel successfully carried out a PCR and gel electrophoresis to identify the microbes found in the DNA from the soil reported yesterday, results are yet to be analyzed. In the afternoon another PCR targeting biogeochemical cycling genes was carried out on the same DNA, currently waiting for the gel to finish running. DNA extractions took place concurrently on soil gathered in today’s EVA.

EVA# 3 crew intended to use Space suits #2, #3, #4, and #6 but ended up using #1, #2, #4, and #6 due to less charge on #3 (5.7V). We figured it was due to the fan being kept ON at low mode after use last time which was consuming the battery. I have made a note of this and will make sure to check for the suits post-EVA and make sure such a mistake does not happen in the future.
No issues with the radios used for EVA #3.

Questions, concerns and requests to Mission Support:
EVA #003 crew exited through the engineering airlock as the main airlock had malfunctioned while opening it. The main airlock was tightly closed due to the windy conditions observed the day before which led to the harness hook being stuck on the inner door handle. The crew tried opening it but it did not open in time and hence the EVA crew had to use the engineering airlock to exit the Hab. The Hab crew had to cut the harness (running through the inner door) because the crew couldn’t open it at all and the main airlock access is needed for the EVAs in the future.
After returning from the EVA, I accessed the situation and thought of two options to address it but I would need your advice on it:
– Harness going around both the outer door handles, pulling in through the inner door, and securing using the hammer as the length of the harness is not enough for the harness hook to lock on the inner door handle (as shown in the pictures attached) OR
– Change the harness to a new one and have a sufficient length for the harness so that the harness hook locks on the handle as before.
